Effect of 2,3,7,8-tetrachlorodibenzo-p-dioxin on the immune system of laboratory animals.

چکیده
2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) is one of the most toxic compounds known. TCDD and other chlorinated dibenzodioxins have been associated with occupational chloracne in workers engaged in the manufacture of technical chlorophenols and their derivatives such as the herbicide 2,4, 5-trichlorophenoxyacetic acid (2,4,5-T) (1), in lethal liver necrosis in rabbits (2), and in the chick edema disease (3). More recently TCDD has been shown to be highly teratogenic (4). Laboratory studies have found TCDD to cause severe atrophy of the thymus at sublethal dose levels (5, 6). Because the thymus is the central lymphoid organ for cellmediated immunity and other environmental chemicals have produced immunosuppression (7), studies to test the immune response in TCDD-treated laboratory animals were conducted. Methods selected to assess cell-mediated immunity were delayed type hypersensitivity to tuberculin in guinea pigs and rats, and a local graft versus host reaction in mice. The effect of TCDD on the humoral immunity was also studied in guinea pigs.

This study assessed the effects of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), 2,3,4,7,8-pentachlorodibenzofuran (PeCDF), and 2,3,7,8-tetrachlorodibenzofuran (TCDF) on the reproductive performance of female mink (Mustela vison) and the viability and growth of their offspring. Nine adult female mink were randomly assigned to one of 13 dietary treatments (one control and four doses each of TCDD, ...
متن کاملToxic effects of 2,3,7,8 tetrachlorodibenzo 1,4 dioxin in laboratory workers.
The toxic effects on three young scientists who had transient minimal exposure to 2,3,7,8 tetrachlorodibenzo 1,4 dioxin (dioxin) are described. Two of them suffered from typical chloracne. Delayed symptoms about two years after initial exposure occurred in two of the scientists. These symptoms included personality changes, other neurological disturbances, and hirsutism. All three scientists wer...
متن کاملEffect of a single oral dose of 2,3,7,8-tetrachlorodibenzo-p-dioxin on immune function in male NC/Nga mice.
Exposure to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) induces immunosuppression in humans and animals. However, the effect of TCDD on Th2-type immune responses such as allergic reactions has been unclear. Using NC/Nga mice that developed atopic dermatitis-like skin lesions with marked elevation in plasma of total IgE when bred under conventional conditions, we investigated the effects of a sin...

We evaluated the effect of chronic exposure to dioxin and dioxin-like compounds on the pancreas in female Harlan Sprague-Dawley rats. This investigation represents part of an ongoing National Toxicology Program initiative to determine the relative potency of chronic toxicity and carcinogenicity of polychlorinated dioxins, furans, and biphenyls. Animals were treated by gavage for up to 2 years w...

Rat small intestinal epithelial cells and human colon adenocarcinoma cells cultured on Matrigel expressed the differentiation specific enzyme, sucrase-isomaltase, as determined by indirect immunofluorescence. Rat small intestinal epithelial cells, rat colonocytes, and human colon adenocarcinoma cells developed an altered morphology when cultured on Matrigel and became apoptotic within 24-48 h. ...
